    Greedy Strategy Works for k-Center Clustering with Outliers and Coreset Construction

    We study the problem of k-center clustering with outliers in arbitrary metrics and Euclidean space. Though a number of methods have been developed in the past decades, it is still quite challenging to design quality guaranteed algorithm with low complexity for this problem. Our idea is inspired by the greedy method, Gonzalez\u27s algorithm, for solving the problem of ordinary k-center clustering. Based on some novel observations, we show that this greedy strategy actually can handle k-center clustering with outliers efficiently, in terms of clustering quality and time complexity. We further show that the greedy approach yields small coreset for the problem in doubling metrics, so as to reduce the time complexity significantly. Our algorithms are easy to implement in practice. We test our method on both synthetic and real datasets. The experimental results suggest that our algorithms can achieve near optimal solutions and yield lower running times comparing with existing methods

    Construction of Evaluation System of Effectiveness of Supply Based on Spatial Perspective

    The requirement of building a higher-level National Fitness Public Service system centering on meeting the needs of the people by the Chinese State Council is to guarantee and satisfy the demand of public through effective supply. However, despite of the clear purpose of the task, there is still a gap between the actual and the expected status of National Fitness Public Service supply. Based on the characteristics of the perspective of space, the purpose of this study is to examine how to measure and achieve the effectiveness of National Fitness Public Service’s supply. By reviewing the literature, relevant results of the performance evaluation system of National Fitness Public Service are sorted to provide theoretical support for the value orientation, index selection and other processes of the evaluation system constructed in this study. Current regulations on National Fitness Public Service in various fields in China are classified, and representative indicators are compared, analyzed, and summarized as the reference basis for evaluation standards. The value orientation of the evaluation system is shown in three levels in the spatial dimension: ensuring spatial justice , adapting to spatial needs and building value consensus . The spatial evaluation system is constructed based on the three-dimensional evaluation dimensions of supply capacity , behavioral demand and The degree of spatial coupling , this evaluation system comprehensively reflects the subjective and objective evaluation results. The study shows that when exploring reasons for insufficient effectiveness, factors should be analyzed from both of supply and demand sides. The evaluation results of the evaluation system are characterized by being Relative and Developmental. The evaluation results should be based on the relative effectiveness results produced by comparison, and absolute judgment should be avoided. The relative evaluation results focus on finding the elements, links and mechanisms with deficiencies, avoid stopping at the judgment of pros and cons, and should promote self-perfection, stimulate self-innovation and finally achieve the goal of self-development

    The Long-Term Effects of Acupuncture on Hippocampal Functional Connectivity in aMCI with Hippocampal Atrophy: A Randomized Longitudinal fMRI Study

    Background. Acupuncture has been used to treat amnestic mild cognitive impairment (aMCI) for many years in China. However, the long-term effects of continuous acupuncture treatment remained unclear. Objective. We aimed to explore the long-term effects of continuous acupuncture treatment on hippocampal functional connectivity (FC) in aMCI. Methods. Fifty healthy control (HC) participants and 28 aMCI patients were recruited for resting-state functional magnetic resonance imaging (fMRI) at baseline. The 28 aMCI patients were then divided into the aMCI acupuncture group, which received acupuncture treatment for 6 months, and the aMCI control group, which received no intervention. All aMCI patients completed the second resting-state fMRI scanning after 6 months of acupuncture treatment. Analysis based on the region of interest and two-way analysis of covariance were both used to explore the long-term effects of acupuncture on cognition change and hippocampal FC in aMCI. Results. Compared to HC, aMCI showed decreased right hippocampal FC with the right inferior/middle temporal gyrus (ITG/MTG), left amygdala, and the right fusiform and increased FC with bilateral caudates at baseline. After acupuncture treatment, the right hippocampal FC with right ITG/MTG enhanced significantly in the aMCI acupuncture group, but continued to decrease in the aMCI control group. Whole brain FC analysis showed enhanced right hippocampal FC with the right ITG and the left MTG in the aMCI acupuncture group relative to the aMCI control group. Furthermore, FC strength of the right hippocampus with right ITG at baseline was negatively correlated with the changes in memory scores of aMCI acupuncture patients. Conclusion. Acupuncture treatment could alleviate the progression of cognitive decline and could enhance hippocampal FC with ITG and MTG in aMCI that may be associated with resilience to resistant against neurodegeneration. The findings provided a better understanding of the long-term effects of acupuncture treatment and confirmed the therapeutic role of acupuncture in aMCI

    Improved Response to nab-Paclitaxel Compared with Cremophor-Solubilized Paclitaxel is Independent of Secreted Protein Acidic and Rich in Cysteine Expression in Non-Small Cell Lung Cancer

    Background:The secreted protein acidic and rich in cysteine (SPARC) is a matricellular glycoprotein that is produced by tumor and/or neighboring stroma. SPARC expression is thought to facilitate the intracellular accumulation of nanoparticle albumin-bound paclitaxel (nab-paclitaxel, abraxane [ABX]). Gene hypermethylation is a common mechanism for loss of SPARC expression in non-small cell lung cancer (NSCLC). We aim to demonstrate the role of SPARC expression as biomarker for treatment selection using ABX in NSCLC and to evaluate the presence of synergistic antitumor effect when a demethylating agent is combined with ABX.Methods:We analyzed the SPARC messenger RNA expression and SPARC gene methylation status in 13 NSCLC cell lines and 22 minimally passaged patient-derived (PD) NSCLC tumors using real-time (RT) polymerase chain reaction. The effect of ABX on tumor growth was compared with cremophor-solubilized paclitaxel (taxol) in severe combined immunodeficiency mice bearing SPARC-positive PD xenografts. The effect of pretreatment with a demethylating agent, 5-Aza-2′-deoxycytidine (DEC) in SPARC-negative tumors was assessed.Results:SPARC expression was weak to absent in 62% of established NSCLC cell lines and 68% of PD NSCLC tumor xenografts. SPARC expression could be up-regulated/restored by DEC treatment in both SPARC-negative cell lines and PD xenografts in vitro and in vivo. ABX demonstrated better antitumor efficacy than equitoxic dose of taxol in SPARC-expressing xenografts and some SPARC-negative xenografts. At equimolar doses in vitro, there was similar increased cytotoxicity on DEC pretreatment with either ABX or taxol in SPARC-negative cell lines. At equitoxic doses, there was similar additive antitumor activity of DEC with either ABX or taxol in SPARC-negative PD xenografts.Conclusion:Endogenous SPARC status is somewhat uncorrelated with response to ABX in NSCLC. The greater antitumor effect of ABX compared with equitoxic dose of taxol observed in SPARC-expressing NSCLC tumors can also be seen in some SPARC-negative tumors.     Commercial carbonate electrolytes with poor oxidation stability and high flammability limit the operating voltage of Li-ion batteries (LIBs) to ~4.3 V. As one of the most promising candidates for electrolyte solvents, sulfolane (SL) has received significant interest because of its wide electrochemical window, low flammability and high dielectric permittivity. Unfortunately, SL-based electrolytes with normal concentrations cannot achieve highly reversible Li+ intercalation/deintercalation in graphite anodes due to an ineffective solid electrolyte interface, thus undermining their potential application in LIBs. Here, a low-concentration SL-based electrolyte (LSLE) is developed for high-voltage graphite||LiNi0.8Co0.1Mn0.1O2 (NCM811) full cells. A highly reversible graphite anode can be achieved through the preferential decomposition of the dual-salt LiDFOB-LiBF4 in the LSLE. The addition of fluorobenzene further restrains the decomposition of SL, endowing uniform, robust and inorganic-rich interphases on the electrode surfaces. As a result, the LSLE with improved thermal stability can support the MCMB||NCM811 full cells at 4.4 V, evidenced by an excellent cycling performance with capacity retentions of 83% after 500 cycles at 25 ℃ and 82% after 400 cycles at 60 ℃. We believe that the design of this fluorobenzene-containing LSLE offers an effective routine for next-generation low-cost and safe electrolytes for high-voltage LIBs

    Abnormal global functional network connectivity and its relationship to medial temporal atrophy in patients with amnestic mild cognitive impairment

    <div><p>Background</p><p>Amnestic mild cognitive impairment (aMCI), which is recently considered as a high risk status for developing Alzheimer’s disease (AD), manifests with gray matter atrophy and increased focal functional activity in the medial temporal lobe (MTL). However, the abnormalities of whole-brain functional network connectivity in aMCI and its relationship to medial temporal atrophy (MTA) remain unknown.</p><p>Methods</p><p>In this study, thirty-six aMCI patients and thirty-five healthy controls (HCs) were recruited. Neuropsychological assessments and MTA visual rating scaling were carried out on all participants. Furthermore, whole brain functional network was constructed at voxel level, and functional connectivity strength (FCS) was computed as the sum of the connections for each node to capture its global integrity. General linear model was used to analyze the FCS values differences between aMCI and HCs. Then, the regions showing significant FCS differences were adopted as the imaging markers for discriminative analysis. Finally, the relationship between FCS values and clinical cognitive scores was correlated in patients with aMCI.</p><p>Results</p><p>Comparing to HCs, aMCI exhibited significant atrophy in the MTL, while higher FCS values within the bilateral MTL regions and orbitofrontal cortices. Notably, the right hippocampus had the highest classification power, with the area under receiver operating characteristics (ROC) curve (AUC) of 0.790 (confidence interval: 0.678, 0.901). Moreover, FCS values of the right hippocampus and the left temporal pole were positively correlated with the cognitive performance in aMCI.</p><p>Conclusion</p><p>This study demonstrated significantly structural atrophy and raised global functional integrity in the MTL, suggesting simultaneous disruption and compensation in prodromal AD. Increased intrinsic functional connectivity in the MTL may have the potential to discriminate subjects with tendency to develop AD.</p></div
